855 Histone deacetylase activity and expression alters with increasing age in primary human keratinocytes

During normal development and throughout life, epigenetic changes arise in response to an individuals’ behavior environment. Histone deacetylases (HDACs) are chromatin-modifying enzymes that remove acetyl groups from histones, inducing conformational DNA changes, so regulating gene expression. To date, four classes of HDACs have been identified: class I (HDACs-1, -2, -3 & -8); II (HDACs-4, -5, -6, -7, -9 -10); III (SIRT1-7) IV (HDAC11), but their role keratinocyte biology is largely unknown. In this study, we investigated the activity abundance both immortalized human keratinocytes (HaCaT) primary epidermal (NHEK) juvenile (n=3, mean age: 6yrs), middle-aged 51yrs) elderly 80yrs) donors. We found HaCaT cells expressed high HDAC when compared (P<0.05) NHEKs; however, was comparable between HaCaTs NHEKs derived Western blot analysis revealed HDACs-1, -5 were at significantly higher levels as juvenile, or NHEK (P<0.05). further lower HDACs-4 -6 than donors (P<0.05); expression HDACs-7, -8, -9, -10 -11 invariant cells. Immunocytochemistry confirmed findings. Taken together, these data suggest cell type-specific cells, alters life course. Further studies exploring consequences such warranted transcriptional translational level better understand contribution aging.
